Annexins are calcium-dependent phospholipid-binding proteins. All Annexins have a putative binding site for protein kinase C. Annexin V also binds to phosphatidylserine (PS), which is located on the inner face of the cellular plasma membrane. However, during apoptosis a cell may translocate PS from the inner face of the plasma membrane to the cell surface. Once on the apoptotic cell surface, PS may be easily detected by the use of Annexin V, which has a strong natural affinity for PS. Thus Annexin V will bind to exposed PS on cells allowing the detection and analysis of apoptosis through flow cytometry or fluorescence microscopy.
